How to Collect Plasma from Whole Blood Before Cell Isolation

Plasma and serum are two important components of whole blood that are widely used in both diagnostic and research applications. Plasma is the pale yellow, extracellular fluid that contains proteins, electrolytes, extracellular vesicles (EVs), and other soluble factors. It provides a cell-free medium rich in biomarkers for downstream analysis. Serum, obtained after blood coagulation, can also be used in many of the same assays.

Collecting plasma enables analysis of biomarkers such as proteins, nucleic acids, and EVs, while also preserving the cellular fraction of whole blood for further processing. Once the plasma is removed and replaced with the appropriate medium, the remaining cells, including peripheral blood mononuclear cells (PBMCs), can be isolated and characterized.

This protocol describes how to collect plasma from a whole blood sample and prepare the cellular fraction for downstream isolation of specific cell populations. Protocol

Part I. Plasma Collection

There are multiple options for generating or sourcing astrocytes:

  1. Collect whole blood using an appropriate anticoagulant.
  2. Centrifuge the blood at 2000 - 2500 脳 g for 10 - 15 minutes at room temperature (15 - 25掳C) with the brake off.
  3. Carefully aspirate the pale yellow plasma layer, leaving ~1 cm above the red blood cell pellet. To minimize disturbance of the cellular components and accidental collection, position the pipette tip at the liquid surface and gradually lower as plasma is collected.
  4. Transfer the collected plasma into a new, labeled collection tube.

Part II. Preparing the Blood Sample for Cell Isolation

  1. To continue with cell isolation from the residual blood sample, replace the removed plasma with an equal volume of the recommended medium. Please refer to the Product Information Sheet for your chosen cell isolation product to determine the appropriate medium.
