Easily isolate highly enriched CD4+CD127low T cells using the RosetteSep⢠Human CD4+CD127low T Cell Enrichment Cocktail. CD127, the α-chain of the IL-7 receptor, plays a critical role in distinguishing human regulatory T cells (Tregs) from activated T cells. While it is broadly expressed on most mature T cells, where it supports proliferation and differentiation, regulatory T cells show little to no CD127 expression. This inverse correlation with FoxP3 makes CD127 an important marker to accurately identify and characterize T cell populations.
This RosetteSep⢠Human CD4+CD127low T Cell Enrichment Cocktail is designed to isolate CD4+CD127low T cells directly from whole blood by negative selection. Unwanted cells are targeted for removal with tetrameric antibody complexes recognizing non-CD4+CD127low T cells and glycophorin A on red blood cells (RBCs). When centrifuged over a buoyant density medium such as ³¢²ā³¾±č³ó“Ē±č°ł±š±čā¢, RosetteSep⢠DM-L Density Medium, or RosetteSep⢠DM-M Density Medium, the unwanted cells pellet along with the RBCs. The purified CD4+CD127low T cells are never labeled with antibodies and are easily collected as a highly enriched population at the interface between the plasma and the buoyant density medium. Isolated cells are immediately available for downstream applications such as flow cytometry, cell culture, or DNA/RNA extraction.
