Overview

Isolate highly purified human lymphoid cells from fresh human whole blood, buffy coat, or leukoreduction system chamber (LRSC; also known as an LRS cone) samples with ease, using immunomagnetic positive selection, wtih the EasySep? HLA Chimerism Whole Blood Lymphoid Positive Selection Kit. Widely used in published research for more than 20 years, EasySep? combines the specificity of monoclonal antibodies with the simplicity of a column-free magnetic system.

In this EasySep? positive selection procedure, desired cells are labeled with antibody complexes recognizing CD3, CD19, and magnetic particles. Labeled cells are separated using an EasySep? magnet and by simply pouring or pipetting off the unwanted cells. The cells of interest remain in the tube. Following magnetic cell isolation, the desired lymphoid cells are ready for downstream applications such as flow cytometry, culture, DNA/RNA extraction, or lineage-specific chimersim analysis. The CD3 antigen is expressed on T lymphocytes and NKT cells while the CD19 antigen is expressed on B cells and follicular dendritic cells.

Data Figures

Starting with human whole blood, the lymphoid cell content (CD3+CD19+) of the isolated fraction is typically 99.5 ± 0.2% (gated on CD45; mean ± SD using “The Big Easy” EasySep? Magnet). In the above example, the purities of the start and final isolated fractions are 16.2% and 98.5%, respectively.
